Well, having just moments ago finished the final episode in Code Geass season 2, I can most certainly and whole-heartedly endorse it as one of the very best animes I have ever watched. Mai Hime, K-On, GSG S1, Last Exile, Noir and TTGL I also recommend. Elfin Lied however, although I most definitely liked it, is not for everyone. It is very bloody bordering on the gratuitous, very heart-wrenching at times and extremely dark in its overall story tone. So if you like that kind of violent, tragic style, then you may like it. If not, than you may want to avoid it.
Oh yeah, there’s also boobs. And loli boobs.

I've seen most of the titles recommended here, and I have to say it is quite the list. I just want to add:

Ga Rei Zero: An thrilling and dark ride propelled by malevolence and despair.

Darker than Black: If you are looking for something that doesn't require a lot of thinking but has gratuitous action, then DtB is the show for you.

it's sort of an old list though I would also suggest Puella Magi Madoka Magica, a new show that isn't finished yet but is probably one of the best I've ever seen. Just research the ones that look interesting to you.

My picks in terms of anime in general...

Macross Frontier: Awesome action, awesome music, and awesome characters. So basically in general... it's awesome.
Mai-HiME: Ending is controversial since a lot of people thought it ruined the show, but a great exercise in long term character development. Starts off light, later gets really dark. Excellent rewatchability due to how many little details early on take on more meaning once you know what happens later.
Full Metal Panic: Mecha action/high school romantic comedy. A socially inept teenage mercenary is assigned to go undercover to protect a short tempered high school girl without her knowledge or permission. More down to earth than it sounds, at least in the serious seasons (the completely comedy oriented second season tends to be more bizarre).
School Rumble: Bit of a mixed bag since it takes a while to get good then starts to suck somewhere in the second season, but between those points this show is flat out hilarious. Very good at thriving on its own insanity.
Ga-Rei Zero: A rather dark and tragic series with excellent characterisation. However it also contains some lighter hearted parts, which I think is part of what makes the darker parts so effective in contrast. Also notable for having a first episode that was essentially an elaborate viral marketing scheme for the rest of the series.

Given what you're looking for School Rumble and maybe Full Metal Panic would probably be your best bets (and much as I tend to favour the more serious FMP series you might find you prefer the more comedic second season).

Only one I've seen in its entirety is UFO Ultramaiden Valkyrie, basically super-ecchi magical girls + harem + aliens/futuristic + neko maids + comedy + romance love triangles + slice of life + light lolicon. It's basically the omni-anime, you'll either continue to watch it out of morbid curiosity or turn it off before the opening ends because your eyes will be bleeding. Gets sort of heavy/dramatic in the second season, but also the best. Good soundtrack, though.

On a related note School Rumble was the first anime I watched years ago, it definitely has "something" that makes it worth watching, I think its mainly the very wide cast of somewhat-interesting characters that keeps it sort of interesting even after the first season. That covers comedy/romance.

For just comedy there's always Pani Poni Dash. It's absolute craziness and insanity about 90% of the time.

Less Serious, slice of life-type comedy stuff

Azumanga Daioh: A classic slice of life comedy revolving around the experiences of a group of high school girls. Includes a number of memorable characters such as the spaced-out Ayumu "Osaka" Kasuga, Chiyo Mihama, a child prodigy who skipped several grades to end up in high school, and Tomo Takino, an overenergetic, but not always overly bright girl who is constantly challenging people and trying to one-up them. I'd highly reccomend it if you haven't seen it already.

Minami-Ke: A comedy/slice of life series about the three Minami sisters, Haruka, the oldest sister, and the kind one, Kana, the middle sister, who is not the brightest light on the proverbial Christmas tree, and Chiaki, youngest sister, who is often rather condescending at at times downright rude.

Will add more later, including some more serious stuff and some less serious, more "fun" action series.

Although I haven't watched all of it, another comedy/rmance/action type anime you might want to look into is To Love Ru. The basic premise is that, an otherwise normal high-school boy, Rito Yūki, who is too shy to confess his feelings for the girl of his dreams, meets an alien princess who drops naked into his bath after running away from a politically marriage arranged by her father. I think that about sums up the tone of the show.

Azumanga Daioh is one of the few Anime I can watch over and over and not go mad. Two thumbs up. d(o_o)b

Strike Witches has great characters too. Like I always say, as long as it has a great plot, characters, and animation I don't care about fanservice. (Not that I enjoy it mind you. Guy fanservice, however? Yes please!) X3

"Best Anime" is such a subjective term, but I will state that there are some recent shows and old favorites I feel are standouts in their particular genres:

Comedy: I found Shinryaku! Ika Musume LOL funny. Older classics include Azumanga Daioh and Strawberry Marshmallow.

Romantic Comedy: Tenchi Muyo (I prefer the English dub), Ah! My Goddess! and anything by Ken Akamatsu (Love Hina, Negima! Magister Negi Magi). Also love the two Spice & Wolf series.

Action/Adventure: Ghost in the Shell: Stand Alone Complex Seasons 1 and 2 (also prefer the English dub), Angel Heart (a City Hunter spinoff) and Gunslinger Girl.

"Slice of Life" (School Edition): K-On! (and to a lesser extent, K-On!!). Hatsukoi Limited was good, IMO, and the first Minami-ke is a winner (the sequels are hit and miss, IMO).

Drama: I'm a huge fan of the anime adaptions of Takako Shimura's works, including Aoi Hana and Hōrō Musuko (Wandering Son). Also loved Katanagatari (even if I raged something fierce with the next-to-last episode) and Puella Magi Madoka Magica is getting better with each episode.

Science Fiction: Both Dennou Coil and Kino's Journey were amazing and I really liked RD Sennou Chosashitsu, RideBack and Macross Frontier.

Guilty Pleasures: The two Rosario+Vampire series and the two Strike Witches series.

If you're looking for a gritty dark comedy/action show Black Lagoon is perhaps one of my favorite anime of all time. Others here would agree with me that it is a fun series.
